-
There is a need to have early clinical and laboratory diagnostics of immunodeficiencies against the background of replicative activity of Epstein-Barr virus in particular in case of atypical or latent clinical course of this infection. It is known that Epstein-Barr virus belongs to immunotropic viruses that can damage not only immune cells but also important organs or systems cells. Expected results: timely diagnostics of replicative form of the infection among pregnant women would prevent pregnancy and birth pathologies, among infants and children it would prevent damage of internals (liver, kidneys, pancreas, gastrointestinal channel), endocrine glands, central and peripheral nervous system, progression of immunodeficiencies, serious post-vaccination accidents, that in its turn will allow to prescribe ethiotropic and pathogenetic treatments for prevention of heavy pathology progression against the background of replicative activity of this infection.

CLUSTER 8: Innovation and Nanotechnologies: In Agriculture, Food Sector and Medicine
The activity of the cluster 8a (M. Spivak) will be aimed at solution of the following main problems:
-
The search for modern reliable and not very expensive methods of plant protection from various infectious agents has in recent years resulted in deeper understanding of necessity to constantly support an organism’s non-specific resistance on the physiological level. This significantly reduces expenses at development and use of specific immune protection methods such as vaccines and serums, decreases the risk of antibiotics resistance development, reduces their side effects, allows to use organism support non-specific methods almost for all strata of population with much lesser expenses on labour and money.
-
One of the world-wide problems of infectious diseases treatment today is not only the rapid spread of “old” infection, emergence of new pathogens mostly of viral nature, but also an unpredictable emergence of aggressive forms of opportunistic commensal organisms. These phenomena in their turn are caused by general loss of organism’s protective forces strengths, spread of immune unbalance states (loss of strength or overexcitement), which are caused by environmental changes, wide use of various chemical preparations, including novel antibiotics and consequent resistance towards them, organism’s normal flora malfunction and similar phenomena. Changes in immunity states are, in their turn, responsible for disposition towards opportunistic infections and oncological diseases (in case of weakened immune system) or towards chronic inflammatory states, autoimmune diseases, oncological diseases, allergic states etc. (in case of overexcited immune system). This created a complicated set of problems related to necessity of finding some novel methods for organism immune status balancing on the level of receptor-ligand interaction during infectious diseases. That is why it is extremely important to develop novel biotechnological preparations – immunobiotics – based on previously selected and characterized strains of lactobacteria and bifidus bacteria with high level capability to induce different nature cytokines and to balance organism’s immune responses during various forms of infectious diseases – bacterial, viral, fungus.
-
It is know that genital herpes virus belongs to viruses able to damage not only immune-competent cells but also main organs and systems cells. Early clinical and laboratory immunological diagnostics of immunodeficient states against the background of the viral infections is needed, especially in case of atypical or latent clinical course of the infection. That is why timely diagnostics of this infection among pregnant women is an extremely important task for perinatal medicine – since this would help to prevent pathologies of pregnancy and birth – and for paediatrics because this would prevent injuries to internals such as liver, kidneys, pancreas, gastrointestinal tract, endocrine glands, central and peripheral nervous system as well emergence of immunodeficient states and serious post-vaccination complications. These factors taken together give possibility to use etiotropic and pathogenetic treatment successfully in order to prevent serious pathologies against background of virus infection replicative activity among children and teenagers.

Research of biological properties of nano-dispersed cerium is one of the priority directions of our studies since such nanos-systems are characterized by specific regulatory properties on the level of living objects and are able to delicately react to changes in cell microenvironment. They demonstrate anti-oxidant and radiation protection properties and, supposedly, antitumour properties. Our research proved that cytotoxicity of nano-particles diminishes in the row “rodent – primates” and that cerium sol activity depends on nano-particles’ size as well as on their super oxiddisemutase properties. We have conducted novel experiments on detection of cerium nano-particles sol activity in cell-cultures. We plan to develop principally new foundations for development of new medicines based on cerium dioxide nano-particles and its compositions with biologically active high-molecular substances, which would be efficient and affordable. Their introduction would improve efficiency of infectious disease prophylaxis and therapy.
-
It is know that enrichment of poultry diet with antioxidants (vitamins C and B, selenium) improves reproductive properties of poultry. Use of nano-crystal cerium dioxide is much more efficient than use of traditional antioxidants – vitamins C, E or melatonin.
-
Industrial aviculture and its peculiarities such as malnutrition, microbial loading, abiotic factors, intensive keeping etc. cause formation of excessive free radicals of oxygen in poultry’s organisms. These radicals damage biologically important molecules (proteins, lipids, DNA) and thus negatively influence reproduction, development and especially immune system of poultry. Imbalance of quails and chickens rations regarding contents of natural antioxidants (vitamins C, E, selenium) activates ferments, in particular kaspazes, responsible for mechanism of programmed cell death in most important – germinal – organs. It is supposed that introduction of such a powerful antioxidant as nano-crystal cerium dioxide to quails and chickens ration with water would allow to increase adaptation capability of the poultry to oxidation stress and to significantly increase its reproductivity.
-
The research on biological properties of gold nano-particles as biosafe and biocompatible nanoconstructions for address delivery of cardiotropic preparations (in particular “Simdax”) is very promising. For this purpose, we developed and implemented original protocols of different size gold nano-particles colloid-chemical synthesis. Using chemical condensation method sphericalgold-nanoparticles of 10, 20, 30 and 45 nm size were synthesized. It was found out that 30 nm gold nano-particles proved to be most biosafe and biocompatible according to all markers, determined during in vitro and in vivo experiments. 30 nm gold nano-particels are not immunetoxic and in case of intravenous introduction to intact rats specifically accumulate in arterioles endotheliocytes and myocardium capillaries that actually suggest their future application as a vector for address delivery of cardiotropic preparations. We have established peculiarities of gold nano-particles conjugation with model proteins. Aggregate stability of gold-nanoparticles in model blood system was also researched. On the basis of cardiotropic preparation “Simdax” and gold nano-particles a new preparation was developed – “Simdax-NZ” which was able to penetrate through membranes and to accumulate within cells; it was biosafe in terms of cytotoxicity and genotoxicity. The bank of newborn babies with congenital heart disease blood serums was created for hypoxemia detection.
